Come up with a list of 12 characters before looking at the questions.
Mine:
1. Skywise - ElfQuest
2. Ford Prefect - The Hitchhiker's Guide to the Galaxy
3. Crow T. Robot - Mystery Science Theater 3000
4. Fred and George Weasley - Harry Potter
5. Arnold Judas Rimmer - Red Dwarf
6. Elrond - The Lord of the Rings
7. Unicorn/Amalthea - The Last Unicorn
8. Sarah - Labyrinth
9. Guinan - Star Trek: The Next Generation

Which would make a better college professor, 6 (Elrond) or 11 (Leela)? What subjects would they teach? Better? Depends. Do you want detailed ancient knowledge and philosophy? Elrond sweeps it. Do you want ass-kicking? Then Leela. (Actually, Elrond was in the Last Alliance... hmm.)
12 (Malkav) sends 8 (Sarah) on a mission. What is it, and does it succeed? Oh, fuck me. Malkav sending anyone "on a mission" can't be good for the general sanity of the world. What the mission specifically is is of course completely inscrutable, but let me just say -- look out, cousin Bowie! Your fancy architecture is about to get even more Escheresque! (And of course it succeeds. How can you not when you see outside of normal reality in the way that Malkav does?)
What is or would be 9(Guinan)'s favorite book? I wonder if she has one in canon. I don't think I could hazard a guess as to this one.
Would it make more sense for 2 (Ford Prefect) to swear fealty to 6 (Elrond), or the other way around? Probably the other way around, although Ford would surely be rather puzzled by the idea. I can't see Ford swearing fealty to anyone.
For some reason, 5 (Arnold Rimmer) is looking for a roommate. Should 5 (Rimmer) share a studio apartment with 9 (Guinan) or with 10 (Kagome)? Should? Guinan. Maybe she could talk some sense into him. Not likely, though, as we all know Rimmer's idea roommate is himself. (Or so he thought, anyway.)
3 (Crow) challenges 10 (Kagome) to a duel. What happens? Inuyasha shows up and kicks Crow's butt while Kagome hangs back shouting "Inuyasha? Inuyasha! Inuyasha..." in various tones of voice. Afterwards she patches Crow up with some of those modern Band-Aids and stuff she always seems to have around, and they eat instant noodles and Inuyasha shouts a lot for everyone to get off his back.
Suggest a title for a story in which 7 (Unicorn) and 12 (Malkav) both attain what they most desire. uh. "Red Bull and Vodka"?
If 7 (Unicorn) visited you for the weekend, how would you get along? Probably not terribly well, actually. She's a little weepy, and I'm no true magician.
What might 10 (Kagome) shout while charging into battle? "I don't think so, demon!" (duh. actually this is more a paraphrase than anything. I couldn't think of something exact off the top of my head.)

Lots of things tend to go your way when you have untold mastery of Dominate. (Two elves and an Antediluvian? wtf? This sounds like the setup for a "walk into a bar" type joke.)
What would 5 (Arnold Rimmer) most likely be arrested for? Murder in the second degree, one thousand, one hundred and sixty-seven counts, as seen in the fourth-season episode "Justice".
What is 6(Elrond)'s secret? Insert slash joke involving Legolas/Aragorn/Boromir/Celeborn/Haldir here.
If 11 (Leela) and 9 (Guinan) were racing to a destination, who would get there first? Hmm. How do the warp capabilities of the Planet Express ship compare to the Enterprise? I'll say Leela, since there are more deus ex machinas on average in a silly half-hour cartoon than there are in a relatively serious hour-long drama.
3 (Crow) and 7 (Unicorn) tell conflicting versions of events. Which is more reliable? Unicorn in terms of purposeful lies; I don't think she knows how (although she could be mistaken). Not that Crow's a baddie either, but he's certainly known to tell a fib or two. If we're talking likelihood of having observed correctly, then Crow, what with his robotic eyeballs and computer memory and all. Just hook him up and play him back!

Meanwhile, 4 (the Weasleys) has (have) kidnapped 12 (Malkav) in an attempt to force their surrender. Following the wise advice of 5 (Rimmer), they seek out 3 (Crow), who gives them what they need to complete their quest. (Scary.)
What title would you give this fic? "Harry Potter and the Quest-a-rama Dwarf Theater 3000: The Next Masquerade Generation; or, the Crossover Fic from Hell"
Name three people on your friends list who might read it. Assuming (as the next question seems to) that I didn't write it? Maybeamberite or arhuaine or blackthornglade or someone would stumble across it and tack it up for laughs, but I really dunno.
Name one person who should write it. Robert Holdstock. I'd like to see that one done a la Mythago Wood and Lavondyss. Ayup yup.
