I don't generally write markup. Here I was considering it because I doubted my ability to draw the little lj dude. (Judge for yourself whether it was good enough; I suppose, like the rest of my writing, if you could understand it, it was good enough.) I do routinely use the <lj user=xxxxx> thing in email, though, since it's a convenient way to refer to someone whom the recipient may not know by any other name.
The specific O.o emoticon here was pretty tongue in cheek, but in general yes, in informal handwriting I will use emoticons, although always upright like the first smiley face. I don't use the whole bizarre net assortment, though; usually just smile, frown/angry, sad, wink, and something indicating confusion.
What I do not do is speak acronyms out loud. If I am "LOLing" then I just do it. Heh.
